    PAL leader training at Bournemouth University: 12 years on and still evolving

    Peer Assisted Learning (PAL) at Bournemouth University (BU) is a peer mentoring scheme that fosters cross-year support between students on the same course. Coordination of PAL, including leader training, is run centrally within Student and Academic Services by the PAL Coordination Team. Successful applicants attend two days of compulsory training in June or September with optional follow up training sessions offered throughout the autumn term. As with other training programmes for peer learning schemes, including Supplemental Instruction (SI), upon which PAL is based (Arendale 1994; Jacobs et al. 2008), the concept of modelling is integral to the training. Trainers employ small group learning techniques and frequently re-direct questions. Leaders can then use these approaches in their own sessions. Crucially, all attendees lead a simulated PAL session. Weekly follow up training is delivered in collaboration with other support staff, providing information on various academic skills, support services and ideas for related PAL sessions. Like PAL itself, leader training has evolved gradually since it began in 2001. Changes include: training on new online community areas on the University's Virtual Learning Environment; streamlining of initial training in response to trainee feedback. However, the overarching principles of the training, established by the founders of the scheme, remain (Capstick et al. 2004). Qualitative feedback from 2011-2012 trainees after completing training, and from a later survey delivered to them towards the end of their role, has further confirmed the continued power of this training while revealing potential ways to strengthen it

    Metagenomic analysis of the turkey gut RNA virus community

    Viral enteric disease is an ongoing economic burden to poultry producers worldwide, and despite considerable research, no single virus has emerged as a likely causative agent and target for prevention and control efforts. Historically, electron microscopy has been used to identify suspect viruses, with many small, round viruses eluding classification based solely on morphology. National and regional surveys using molecular diagnostics have revealed that suspect viruses continuously circulate in United States poultry, with many viruses appearing concomitantly and in healthy birds. High-throughput nucleic acid pyrosequencing is a powerful diagnostic technology capable of determining the full genomic repertoire present in a complex environmental sample. We utilized the Roche/454 Life Sciences GS-FLX platform to compile an RNA virus metagenome from turkey flocks experiencing enteric disease. This approach yielded numerous sequences homologous to viruses in the BLAST nr protein database, many of which have not been described in turkeys. Our analysis of this turkey gut RNA metagenome focuses in particular on the turkey-origin members of the Picornavirales, the Caliciviridae, and the turkey Picobirnaviruses

    Migratory insights from singing humpback whales recorded around central New Zealand

In the western South Pacific, migratory routes of humpback whales between breeding and feeding areas are unclear. Male humpback whales sing a population-specific song, which can be used to match singers on migration to a breeding population. To investigate migratory routes and breeding area connections, passive acoustic recorders were deployed in the central New Zealand migratory corridor (2016); recorded humpback whale song was compared to song from the closest breeding populations of East Australia and New Caledonia (2015-2017). Singing northbound whales migrated past New Zealand from June to August via the east coast of the South Island and Cook Strait. Few song detections were made along the east coast of the North Island. New Zealand song matched New Caledonia song, suggesting a migratory destination, but connectivity to East Australia could not be ruled out. Two song types were present in New Zealand, illustrating the potential for easterly song transmission from East Australia to New Caledonia in this shared migratory corridor. This study enhances our understanding of western South Pacific humpback whale breeding population connectivity, and provides novel insights into the dynamic transmission of song culture.Publisher PDFPeer reviewe

    Vacuum-assisted sorbent extraction: An analytical methodology for the determination of ultraviolet filters in environmental samples

    Vacuum-assisted sorbent extraction (VASE) has been applied for the first time in the determination of UV filters in water samples in combination with gas chromatography-mass spectrometry. VASE is a variant of headspace extraction which was developed in conjunction with the sorbent pen (SP) technology. This technique combines the advantages of both stir-bar assisted extraction and headspace solid-phase microextraction. The SP traps allowed both reduced pressure in-vial extraction and direct thermal desorption via a unique gas chromatographic injection port. The main parameters that affect the performance of VASE, including both extraction and desorption conditions, were extensively optimized. Under optimum conditions, extraction required 10 mL of sample within 40 mL vials, pH 3.5, ∼30 s of air-evacuation, 14 h incubation at 70 °C, stirring at 200 rpm, and a final water management step conducted at ∼ −17 °C for 15 min. Optimal thermal desorption required preheating at 260 °C for 2 min followed by desorption at 300 °C for 2 min. The beneficial effect of reduced-pressure extraction was demonstrated by comparing the UV filter extraction time profiles collected using VASE to an analogous atmospheric pressure procedure, resulting in up to a 3-fold improvement under optimized conditions. The VASE methodology enabled simultaneous extractions using different SPs without compromising the method reproducibility, which increases the overall sample throughput. The method was characterized by low limits of detection, from 0.5 to 80 ng L−1, and adequate reproducibility, with inter-SP and inter-day relative standard deviation lower than 14%. Tap and lake water was successfully analyzed with the proposed methodology, resulting in relative recoveries of spiked samples ranging between 70.0 and 120%.This is a manuscript of an article published as Trujillo-Rodríguez, María J., Jared L.     Clinical and genetic characteristics of late-onset Huntington's disease

    Background: The frequency of late-onset Huntington's disease (&gt;59 years) is assumed to be low and the clinical course milder. However, previous literature on late-onset disease is scarce and inconclusive. Objective: Our aim is to study clinical characteristics of late-onset compared to common-onset HD patients in a large cohort of HD patients from the Registry database. Methods: Participants with late- and common-onset (30–50 years)were compared for first clinical symptoms, disease progression, CAG repeat size and family history. Participants with a missing CAG repeat size, a repeat size of ≤35 or a UHDRS motor score of ≤5 were excluded. Results: Of 6007 eligible participants, 687 had late-onset (11.4%) and 3216 (53.5%) common-onset HD. Late-onset (n = 577) had significantly more gait and balance problems as first symptom compared to common-onset (n = 2408) (P &lt;.001). Overall motor and cognitive performance (P &lt;.001) were worse, however only disease motor progression was slower (coefficient, −0.58; SE 0.16; P &lt;.001) compared to the common-onset group. Repeat size was significantly lower in the late-onset (n = 40.8; SD 1.6) compared to common-onset (n = 44.4; SD 2.8) (P &lt;.001). Fewer late-onset patients (n = 451) had a positive family history compared to common-onset (n = 2940) (P &lt;.001). Conclusions: Late-onset patients present more frequently with gait and balance problems as first symptom, and disease progression is not milder compared to common-onset HD patients apart from motor progression. The family history is likely to be negative, which might make diagnosing HD more difficult in this population. However, the balance and gait problems might be helpful in diagnosing HD in elderly patients
